Ellagic Acid is a nutraceutical, naturally occurring phenolic compound that has very effective and potent anti-oxidant properties. This compound is especially concentrated in raspberries, grapes, and nuts. It is also present in strawberries, pomegranates, and cranberries. Several studies have found that it may possess cancer-fighting potential, making it a potent anti-carcinogen. Ellagic Acid has the ability to prevent mutations within a cell's DNA. Among the pioneer published research on ellagic acid started in the 1970's and 80's, with the first studies beginning in 1993. These years of extensive studies have yielded in a dietary supplement that is safe, inexpensive, and free of adverse side effects. Nine years of research have demonstrated that a natural product called ellagic acid is causing G-arrest within 48 hours (inhibiting and stopping mitosis-cancer cell division), and apoptosis (normal cell death) within 72 hours, for breast, pancreas, esophageal, skin, colon and prostate cancer cells.

Ellagic Acid supplementation during chemotherapy cancer treatment has been proven in at least one recent study by Italian researchers to significantly reduce side effects among a group of men suffering from advanced prostate cancer (see: Falsaperla M, Morgia G, Tartarone A, Ardito R, Romano G. Ellagic acid and quercetin interact synergistically with resveratrol in the induction of apoptosis and cause transient cell cycle arrest in human leukemia cells
Anticarcinogenic effects of polyphenolic compounds in fruits and vegetables are well established. Although polyphenols naturally occur as combinations, little information is available regarding possible synergistic or antagonistic biochemical interactions between compounds. Identifying potential interactions between polyphenols may provide information regarding the efficiency of polyphenol-containing foods in cancer prevention. The objective of this study was to investigate the interactions of ellagic acid and quercetin with resveratrol, polyphenols which occur in muscadine grapes, with the hypothesis that the selected polyphenols would interact synergistically in the induction of apoptosis and reduction of cell growth in human leukemia cells (MOLT-4). Results indicate that the anticarcinogenic potential of foods containing polyphenols may not be based on the effects of individual compounds, but may involve a synergistic enhancement of the anticancer effects.

In vitro anti-proliferative activities of ellagic acid
The potential cytotoxic and anti-proliferative activities of ellagic acid was evaluated using human umbilical vein endothelial cells (HUVEC), normal human lung fibroblast cells HEL 299, Caco-2 colon, MCF-7 breast, Hs 578T breast, and DU 145 human prostatic cancer cells. Ellagic acid at concentration in the range 10-100 micromol/L did not affect the viability of normal fibroblast cells during a 24-hour incubation. An increase in adenosine triphosphate (ATP) bioluminescence of approximately 18-21% was observed in normal cells incubated with ellagic acid. In contrast, ellagic acid at 1-100 micromol/L dose-dependently inhibited HUVEC tube formation and proliferation on a reconstituted extracellular matrix and showed strong anti-proliferative activity against the colon, breast, and prostatic cancer cell lines investigated. Ellagic acid induced cancer cell death by apoptosis. It was also observed that the mechanism of apoptosis induction in ellagic acid-treated cancer cells was associated with decreased ATP production, which is crucial for the viability of cancer cells.

Pomegranate extracts potently suppress proliferation, xenograft growth, and invasion of human prostate cancer cells
We completed a multicenter study of the effects of pomegranate cold-pressed (Oil) or supercritical CO(2)-extracted (S) seed oil, fermented juice polyphenols (W), and pericarp polyphenols (P) on human prostate cancer cell xenograft growth in vivo, and/or proliferation, cell cycle distribution, apoptosis, gene expression, and invasion across Matrigel, in vitro. Overall, this study demonstrates significant antitumor activity of pomegranate-derived materials against human prostate cancer.

Pomegranate extract improves a depressive state and bone properties in menopausal syndrome model ovariectomized mice
Pomegranate is known to contain estrogens (estradiol, estrone, and estriol) and show estrogenic activities in mice. In this study, we investigated whether pomegranate extract is effective on experimental menopausal syndrome in ovariectomized mice. Overall data suggest that the pomegranate extract inhibits ovariectomy-stimulated bone turnover. It is thus conceivable that pomegranate is clinically effective on a depressive state and bone loss in menopausal syndrome in women.

Chemopreventive effects of pomegranate seed oil on skin tumor development in CD1 mice
Pomegranate seed oil was investigated for possible skin cancer chemopreventive efficacy in mice. In the main experiment, two groups consisting each of 30, 4-5-week-old, female CD(1) mice were used. Overall data showed Pomegrante seed oil (5%) significantly decreased tumor incidence, multiplicity, and TPA-induced ODC activity. Overall, the results highlight the potential of pomegranate seed oil as a safe and effective chemopreventive agent against skin cancer.

Pomegranate juice flavonoids inhibit low-density lipoprotein oxidation and cardiovascular diseases: studies in atherosclerotic mice and in humans
The beneficial health effects attributed to the consumption of fruit and vegetables are related, at least in part, to their antioxidant activity. Of special interest is the inverse relationship between the intake of dietary nutrients rich in polyphenols and cardiovascular diseases. This effect is attributed to polyphenols' ability to inhibit low-density lipoprotein (LDL) oxidation, macrophage foam cell formation and atherosclerosis. Pomegranate polyphenols can protect LDL against cell-mediated oxidation via two pathways, including either direct interaction of the polyphenols with the lipoprotein and/or an indirect effect through accumulation of polyphenols in arterial macrophages. Dietary supplementation of polyphenol-rich pomegranate juice to atherosclerotic mice significantly inhibited the development of atherosclerotic lesions and this may be attributed to the protection of LDL against oxidation.

Pomegranate fruits are widely consumed as juice, and its antioxidant and anti-atherosclerotic activities are attributed to ellagitannin and ellagic acid. Ellagic acid, totol pomegranate tannin and punicalagin but not pomegranate juice could induce apoptosis in colon cancer cells HCT116. However, pomegranate juice had a better antioxidant activity than ellagic acid.

Combined treatment of tumor with ellagic acid and radiation enhances oxidative stress and cytotoxicity in tumor cells in a study of mice. Ellagic acid generated ROS in tumor cells, which increased, by an order of magnitude when cells were treated with ellagic acid in combination with gamma radiation. Ellagic acid protects normal cells against radiation damage.
